Bats carry genetically diverse SARS-related coronaviruses (SARSr-CoVs), some of which can infect humans but not wild-type mice. Obtaining the mouse-adapted rRsSHC014S strain and naming it SMA1901 after isolating the virus from lung homogenate
An ideal animal infection model should be able to reproduce the viral replication levels, clinical features, and pathological features of human patients.
To generate mouse-adapted strains, the researchers continuously transmitted bat-derived coronavirus rRsSHC014S to young adult BALB/C mice via intranasal inoculation. Nineteen generations after inoculation, the infected mice died of severe lung lesions. The researchers isolated the virus from lung homogenate and named it SMA1901 for further identification.
Compared with WT-rRsSHC014S, SMA1901 has 10 non-synonymous mutations and 2 synonymous mutations. Most notably, two mutations, H492Y and L504P, are located in the RBD of the S protein.

Studies have found that although mouse-adapted SARS-CoV-2 strains have increased affinity for the S protein of mouse mACE2, their affinity for human hACE2 remains unchanged, slightly weakened, or enhanced.

Monoclonal antibody SRB12 protects mice from SMA1901 infection
The S protein of SMA1901 is similar to WIV1 in its N-terminal domain, but has a unique RBD epitope.
Studies have found that many SARS-CoV and SARS-CoV-2 neutralizing antibodies and sera are ineffective in inhibiting SMA1901 infection. In addition, the SARS-CoV-2 inactivated vaccine only provided partial protection against rRsSHC014S infection in hACE2 transgenic mice. But in this study, the researchers demonstrated that prophylactic injection of the monoclonal antibody SRB12 effectively protected 6-month-old mice from SMA1901 infection.
It is worth noting that surveillance studies on bat coronaviruses have shown that SARSr-CoV, which has a similar RBD to RsSHC014, is circulating in bat colonies. This indicates that it is necessary to prepare preventive or therapeutic drugs such as SRB12 that are more specific for RsSHC014-related viruses to prevent the emergence of future SARSr-CoV epidemics.

Monupiravir alleviates SMA1901 infection in aged mice
Researchers confirmed that molnupiravir can inhibit the replication of SMA1901. This result was expected since monupiravir targets RNA-dependent RNA polymerase, which is highly conserved among coronaviruses.
Other antiviral drugs targeting SARSr-CoV-2, such as Paxlovid, which targets the major protease (Mpro), have also been shown to be effective in inhibiting SARS-CoV-2 and human coronaviruses OC43 and 229E. Since SMA1901 and some other SARSr-CoV share the same Mpro amino acid residues targeted by Paxlovid as SARS-CoV-2, the researchers speculated that it could also provide cross-protection to SMA1901.
Therefore, the SMA1901 strain provides a useful tool for testing antiviral drugs targeting the conserved replication domain of SARSr-CoV.

SMA1901 does not increase infectivity to humans
The full-length RsSHC014 virus replicates less efficiently in human respiratory tract cells and mice than SARS-CoV virus. In terms of lethality, viral load, and pathogenicity in mice of different ages, the pathogenicity of the mouse-adapted strain SMA1901 is lower than that of SARS-CoV.
In addition, in vitro infection results showed that the replication efficiency of SMA1901 in human cell lines was comparable to that of WT-rRsSHC014S, indicating that the infectivity of the mouse-adapted strain to humans is not enhanced.
In conclusion, the mouse-adapted bat SARSr-CoV strain SMA1901 established in this study can induce age-related coronavirus pneumonia symptoms in BALB/c mice and can also well reproduce the clinical characteristics of human patients. Therefore, this model is of high value for studying the pathogenesis of bat SARSr-CoV and evaluating vaccines and antiviral drugs.
It is worth noting that SMA1901 and WT-rRsSHC014S are considered to be viruses of low pathogenicity to humans. Given the ongoing genetic recombination and mutation of coronaviruses occurring naturally in bat populations, as well as their potential for spillover and adaptation in other species, continued monitoring of this class of viruses in bats and other wildlife is warranted.
