Research reveals the molecular mechanism of the elderly susceptible to SARS-CoV-2

Liu Guanghui, a researcher at the Institute of Zoology, Chinese Academy of Sciences, and his collaborators used cynomolgus monkeys as the research object to draw the single-cell nuclear transcriptome map of young and old non-human primate heart and lung tissues for the first time, revealing that the SARS-CoV-2 receptor ACE2 plays a role in aging. The up-regulated expression in the long-type cardiopulmonary system is one of the potential mechanisms of elderly individuals susceptible to the novel coronavirus, and clarifies the key role of the cytokine IL7 in mediating the up-regulation of aging-related ACE2. Relevant research results were published in "Cell Research" on 7 September.

Based on the data analysis of the single-cell atlas, the researchers found that increased cell-specific inflammation and impaired virus defense are important features of non-human primate cardiopulmonary aging. On the one hand, inflammation-related cell types such as mast cells, plasma cells, and CD8+T cells accumulate in the lung tissue of elderly individuals; on the other hand, the aging of the cardiopulmonary system (especially lung tissue) is manifested by excessive resident immune cells Activation and enhanced inflammatory interaction between immune cells and stromal cells.

Zhang Weiqi, the co-corresponding author of this article and a researcher at the Beijing Institute of Genomics of the Chinese Academy of Sciences, said, "As age increases, the number of positive cells for the covi receptor ACE2 or the expression of ACE2 increases in alveolar epithelial cells, cardiomyocytes, and vascular endothelial cells. These findings provide a scientific basis for the increase in susceptibility to the novel coronavirus accompanied by aging."

These findings provide potential molecular targets for enhancing the resistance of elderly individuals to covid and intervention strategies."
